胰岛素(insulin)是胰腺中胰岛β细胞分泌的一种双链形式 ( α, β) 的多肽激素 ,由 51个氨基酸组成 ,其中α链包含 21 个 氨基酸 ,β链包含30 个氨基酸。两条链间通过二对二硫键连接 ,而α链自身还含有一对二硫键。胰岛素的分泌过程是:前胰 岛素原的氨基末端携带有一段 23-30 氨基酸的信号肽 ,该信号肽可将前胰岛素原转移至内质网 ,然后立即被切断 ,释放出 胰岛素原。胰岛素原由胰岛素β链、C-肽和α链组成 ,释放出的胰岛素原随即被转至高尔基体 ,在此被包装并转化成胰岛素,与 C-肽一同释放至血液。胰岛素是目前已知的机体内唯一能降低血糖的蛋白激素 ,不仅负责调控细胞对葡萄糖、氨基酸和 脂肪酸的摄入 ,利用和储存 ,而且抑制糖原 ,蛋白质和脂肪的降解发生。
本品为大肠杆菌表达的全长序列的人胰岛素,分子量大约为 5800 Da,结构与天然来源的人胰岛素相同,常用作细胞培养的辅助成分 ,常见使用浓度为 1~10 μg/mL。

使用说明
重组人胰岛素以粉末状态供应,一般用稀盐酸溶液(10mM)溶解制成每1ml中含1~2mg的储备液,然后除菌过滤,滤液分装后可在-20℃条件下保存6个月。应避免反复冻融。
冻干粉-25~-15℃保存 ,有效期 3年。储存液-25~-15℃保存 ,有效期 6 个月。
Q:能否做活体动物实验?能否观察胰岛素耐受量?
A:不能做活体实验,人的细胞可以观察胰岛素耐受。
Q:这个有配套的缓冲液吗?
A:没有缓冲液,按照说明书上用稀醋酸或者稀盐酸配置成母液就可以了。
Q:这个做脂肪细胞诱导可以吗?
A:文献中是有以胰岛素诱导脂肪细胞的,不过咱的产品没有客户验证过。这个和胰岛素作用是一样的,理论上是可以的,可以让客户尝试一下。
